Proteolysis is one of the key techniques of proteomics research.The development of immobilized enzyme reactors (IMER) enables the proteolysis fast and efficient.Studies show that kinds of hydrophilic monolithic supports can reduce some undesired nonspecific adsorption of proteins and peptides on IMER.In our studies, with acrylamide as the hydrophilic monomer and N,N_-methylenebisacrylamide as the cross-linker, a completely hydrophilic supports was prepared in capillary by free radical polymerization.Reports also showed that the efficiency of proteolysis could be accelerated with the aid of microwave, ultrasonic waves, infrared or laser irradiation, and so on.Herein, a continuous laser radiation (808 nm with a power of 5 W) was used to irradiate the IMER perpendicularly for enhancing proteolysis.
