Objective:To observe the effect of transplantation of Schwann cells and neural stem cells embedded to the chitosan-collagen complex film to the spinal cord injury site in Wistar rats.Method:Schwann cells from the sciatic nerve of the Wistar rat were isolated and cultured by primary cell culture and purified by rapid trysinization and different velocity of adhesion to the bottom of culturing bottle.These Schwann cells were distinguished by anti S-100 protein IgG after cultured for four generation.A neural stem cell line which had ability to form clones was isolated from the brain of Wistar rat embryo by serum-flee cell culture.The clone was passaged continuously for 4 generation in order to acquire a large amount of clones,and indirect immunofluorescence was used to detected Nestin antigen.The two type of cells were embedded to the chitosan-collagen complex film and analysed the compatibility between them.
