It is widely accepted that a battery of in vitro and in vivo tests is utilized to the potential test the Mutagenicity and Carcinogenicity of a test substance.However, the false positive and negative results are difficult to identify via effective and globally harmonized principle and methods.As to mutagenicity testing, a tiered approach has been performed globally.However, at the in vitro screening stage, the high frequency of false positive/negative results is a common issue of these screening tests.Test methods and culture conditions can affect the in vitro test results of bacteria and mammalian cells.Here we, through a case study, access the indirect mutagenicity by simulating the biotransformation and degradation of the parent compounds, therefore identify the in vitro results as false positive.
