Microtubule-associated protein tau (tau protein) is a protein mainly expressed in neuronal tissues.It binds to tubulin,a main component of cytoskeleton,promotes its polymerization and stabilizes microtubule.Generally,phosphorylation attenuates tau protein to bind microtubule,then tau protein polymerizes itself,forms aggregations and intracellular inclusions.Aggregated tau protein is thought to impair axonal transport and neuronal cell functions,which leads to cell death.Also mutated tau proteins can form insoluble aggregations and depositions.Neurodegenerative disorders that show tau protein depositions are called tauopathies.Tauopathies have broad spectrum of neurodegenerative diseases,including Alzheimer s disease (AD),Pick s disease,progressive supranuclear palsy (PSP),corticobasal degeneration (CBD),frontotemporal dementia and parkinsonism linked to chromosome 17 (FTDP-17),or Amyotrophic Lateral Sclerosis-parkinsonism-dementia complex (ALS/PDC).There have not been established causative treatments for tauopathies,although tauopathies tend to show more disastrous clinical courses.
