MUC1在肿瘤发生与治疗中的作用

MUC1是一种广泛表达于呼吸系统、消化系统和生殖系统等分泌型上皮细胞管腔表面的高分子量糖蛋白.临床研究表明MUC1在70%肺癌,结肠癌等实体瘤中呈50-100倍高表达,并伴随着极性消失.该现象尤其存在于95%的乳腺癌中.MUC1异常表达与肿瘤的恶性程度及预后不良密切相关. 本课题组围绕MUC1在肿瘤发生、转移及治疗中的作用进行了较为系统的研究,前期结果发现:(1)建立了MMTV-MUC1-CD过表达小鼠模型,进一步从整体水平阐明了MUC1-CD激活Wnt信号通路,导致乳腺增生的分子机制;(2)在食管癌中阐明了MUC1通过上调MMP13,增强细胞移行、浸润及与细胞外基质作用能力;(3)阐明了MUC1与DNA损伤修复的关系,以及过度表达的MUC1引起放疗不敏感的分子机制,为靶向MUC1的肿瘤生物治疗与化学治疗相结合提供了理论基础。利用小细胞肺癌细胞系研究发现抑制MUC1表达的使细胞对紫杉醇更敏感;对细胞周期的分析发现,MUC1能抵抗紫杉醇诱导的细胞周期阻滞及细胞内Cyclin B1的累积;上述结果提示MUC1在细胞分裂中期的调控中发挥作用。
