Free amino acids contents are very important information for studying the genotype, grow condition and environment of plants, and have been detected with many different methods.Here a profiling method with reverse phase-ultra high performance liquid chromatography (UHPLC) coupled with quadrupole time of flight (qTOF) mass spectrometry after 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) derivation was applied and evaluated to qualify and quantify free amino acids in rice grains.28 kinds of amino acids have been identified and 21 of them were quantified simultaneously.Evaluated result showed that quantified 21 kinds of amino acids have good calibration range, a recovery between 67.93%-118.17%, intra repetition 3.16%-19.6%(n=5), inter repetitionl.05%-19.53% (n=4).
