Toll like receptor 4 (TLR4) as pattern recognition receptors plays a key role both in innate and acquired immune response [1].The most important and best characterized ligand for TLR4 is exogenous lipopolysaccharide [2], via serum and cell-surface proteins including lipopolysaccharide binding protein (LBP) and CD14, adapts to the TLR4/ myeloid differentiation 2 (MD2) signaling complex [3].TLR4 is strictly dependent upon the presence of MD2 [4].The TLR4-MD2 signaling complex is an important therapeutic target, not only in infectious diseases but also in other diseases with an inflammatory aetiology, such as cancer, atherosclerosis, asthma and autoimmune conditions.Therefore, TLR4 is a potential drug target for screening compounds with immunomodulatory activity.Compounds that target the interaction between lipid A and TLR4 in the host could lead to the development of new active compounds for use in infectious and non-infectious diseases [6].
