An ultra fast liquid chromatography/ion trap-time of flight mass spectrometry (UFLC/MS-IT-TOF) based serum lipidomics platform was developed to characterize the serum lipid profile of patients with Chronic Hepatitis B (CHB), Cirrhosis, Hepatocellular Carcinoma (HCC) and normal controls.CH3OH/CH2Cl2/H2O solvent mixture was employed to extract lipids from serum, which was simple and reproducible.Reversed-phase liquid chromatography was used to separate the lipids according to their hydrophobicity, possessing the advantage of retention time stability than normal-phase liquid chromatography.Detection was performed on Ion Trap-Time of Flight Mass Spectrometry, which provided high sensitivity to detect low-content lipid component and high accurate m/z to facilitate lipid identification.
