MCM,composed of six polypeptides of related structures,is a central component for the replicative helicase in eukaryotic DNA replication.We previously reported that the recombinant Mcm4/6/7 complex has an intrinsic DNA unwinding activity and each subunit contributes to the helicase activity of the complex by playing distinct biochemical functions (You et al.,MCB 1999; JBC 2002).We have shown that thymine-rich single-stranded DNA specifically stimulates the helicase activity of theMcm4/6/7 complex.The thymine-rich strands derived from human replication origins also served as potent activators for the MCM helicase.
