目的:研究壮骨镇痛胶囊对高转移人乳腺癌MDA-MB-231细胞迁移及其运动信号通路关键蛋白p-JNK和Rac-1表达水平的影响。方法:采用划痕法测定高(5.0%)、中(1.25%)、低(0.63%)不同浓度壮骨镇痛胶囊含药血浆对MDA-MB-231细胞迁移的影响,采用免疫细胞化学法检测不同浓度壮骨镇痛胶囊对MDA-MB-231细胞p-JNK和Rac-1蛋白表达水平的影响。结果:高、中浓度壮骨镇痛胶囊含药血浆显著抑制了乳腺癌MDA-MB-231细胞的迁移和p-JNK、Rac-1蛋白表达,低浓度血浆对细胞迁移和p-JNK、Rac-1蛋白表达无显著影响。结论:壮骨镇痛胶囊可通过改变运动信号通路关键蛋白p-JNK和Rac-1的表达来抑制乳腺癌MDA-MB-231细胞的迁移,且该作用与浓度密切相关。
Objective: To study the effect of Zhuangguizhuangtong Capsule on the migration of high metastatic human breast cancer MDA-MB-231 cells and the expression of key proteins p-JNK and Rac-1 in their motor signaling pathways. Methods: The migration of MDA-MB-231 cells in different concentrations of Zhuangguizeng Capsules (5.0%, middle (1.25%), low The effect of ZhuangzhuoTeng capsule with different concentrations on p-JNK and Rac-1 protein expression in MDA-MB-231 cells was detected. RESULTS: The drug-containing plasma of high and medium concentration Zhuangguizengtong capsules significantly inhibited the migration of breast cancer MDA-MB-231 cells and the expression of p-JNK and Rac-1 protein, the low concentration of plasma on cell migration and p-JNK, Rac -1 protein expression had no significant effect. Conclusion: Zhuangguizhuang Capsule can inhibit the migration of breast cancer MDA-MB-231 cells by changing the expression of p-JNK and Rac-1, a key protein in motor signaling pathway, and the effect is closely related to the concentration.
