目的:探讨肝癌细胞增殖与血清甲胎蛋白(AFP)、癌胚抗原(CEA)水平的关系。方法:抽取2018年1月至2021年1月河南省人民医院收治的80例肝癌患者为研究组,选择同期来院体检的36例健康者作为对照组。采集所有受试者血清,以电化学发光法检测其AFP、CEA水平及细胞增殖分子[高尔基体糖蛋白73(GP73)、磷脂酰肌醇蛋白聚糖3(GPC3)、Dickkopf同源物1(DKK1)、超氧化物歧化酶(SOD)]水平,采集研究组患者肝癌病灶、癌旁病灶标本,并采用酶联免疫吸附法检测细胞增殖分子[DNA甲基转移酶3B(DNMT3B)、斯钙素2 (STC2)、沉默信息调节因子2相关酶6(SIRT6)、亮氨酸拉链EF-hand结构域跨膜蛋白1(LETM1)、EphB4]含量。采用Pearson相关性分析法分析血清AFP、CEA水平与细胞增殖分子表达量的相关性。结果:研究组血清AFP、CEA水平均高于对照组(n P均<0.05);研究组血清GPC3、GP73、DKK1水平均高于对照组(n P均<0.05),SOD水平低于对照组(n P<0.05);肝癌病灶中DNMT3B、STC2、SIRT6、LETM1、EphB4表达量均高于癌旁病灶(n P均<0.05)。Pearson相关分析结果显示,细胞增殖分子GPC3、GP73、DKK1、DNMT3B、STC2、SIRT6、LETM1、EphB4的表达量与血清AFP、CEA水平呈正相关(n r=0.537、0.432、0.383、0.364、0.614、0.571、0.538、0.676,n r=0.439、0.496、0.553、0.468、0.472、0.647、0.391、0.627,n P均<0.05),SOD水平与AFP、CEA水平呈负相关(n r=-0.319、-0.408,n P均<0.05)。n 结论:肝癌患者血清AFP、CEA水平与细胞增殖分子GPC3、GP73、DKK1、DNMT3B、STC2、SIRT6、LETM1、EphB4的表达量呈正相关,与SOD呈负相关,可通过监测患者血清中AFP、CEA水平来判断肝癌的发生、进展程度及评估疗效。“,”Objective:To investigate the relationship between the cell proliferation and the levels of serum alpha-fetoprotein (AFP) and carcinoembryonic antigen (CEA) in patients with liver cancer.Methods:Eighty liver cancer patients admitted to Henan Provincial People’s Hospital from January 2018 to January 2021 were selected as the study group, and 36 healthy patients who came to the hospital for physical examination during the same period were selected as the control group. The serum of all subjects were collected, and the levels of AFP, CEA and the cell proliferation molecules [ Golgi glycoprotein 73 (GP73), Glypican 3 (GPC3), Dickkopf-1 (DKK1) and superoxide dismutase (SOD)] were detected by electrochemiluminescence. The specimens of liver cancer lesions and adjacent lesions in the study group were collected, and the content of the cell proliferation molecules [DNA methyltransferase 3B (DNMT3B), stanniocalcin 2 (STC2), silent information regular factor 2 related enzyme 6 (SIRT6), leucine-zipper-EF hand-containing transmembrane protein 1 (LETM1), EphB4] was detected by enzyme-linked immunosorbent assay. Pearson correlation analysis method was used to analyze the correlation of serum levels of AFP and CEA with the expression of cell proliferation molecules.Results:The levels of serum AFP and CEA in the study group were higher than those in the control group (all n P<0.05). The levels of serum GPC3, GP73, and DKK1 in the study group were higher than those in the control group (alln P<0.05), the SOD level of the study group was lower than that of the control group (n P<0.05). The expression levels of DNMT3B、STC2、SIRT6、LETM1、EphB4 in liver cancer lesions were higher than those in adjacent lesions (alln P<0.05). Pearson correlation analysis showed that the expression of cell proliferation molecules (GPC3, GP73, DKK1, DNMT3B, STC2, SIRT6, LETM1, EphB4) were positively correlated with the levels of AFP and CEA(n r=0.537, 0.432, 0.383, 0.364, 0.614, 0.571, 0.538, 0.676; n r=0.439, 0.496, 0.553, 0.468, 0.472, 0.647, 0.391, 0.627; all n P<0.05), while SOD levels was negatively correlated with levels of serum AFP and CEA (n r=-0.319, -0.408; all n P<0.05).n Conclusions:Serum AFP and CEA levels in patients with liver cancer are positively correlated with the expression of cell proliferation molecules GPC3, GP73, DKK1, DNMT3B, STC2, SIRT6, LETM1, EphB4, and negatively correlated with SOD. The occurrence, progression and curative effect of liver cancer can be judged by monitoring the levels of serum AFP and CEA.
