目的应用生物信息软件分析细粒棘球蚴14-3-3(Eg14-3-3)蛋白氨基酸序列、二级结构、三级结构并预测B细胞表位和T细胞表位,为后续确定和筛选优势抗原表位、研制安全高效的多肽疫苗及特异性诊断抗原奠定基础。方法采用DNAstar软件、ProPred MHC Class-ⅡBinding Peptide Prediction Server和Biosun等软件对Eg14-3-3的序列、结构及B细胞表位和T细胞表位进行预测。结果Eg14-3-3为相对分子质量27 940、含有247个氨基酸的酸性蛋白质,其二级结构主要由有6个α-helix结构中间连接短β-折叠及不规则卷曲结构,序列具有高度多变性,预测该蛋白存在14个B细胞表位区域和6个T细胞表位。结论分析了蛋白氨基酸序列、二级结构、三级结构,并预测出Eg14-3-3存在14个B细胞表位和6个T细胞表位。
Objective To analyze the amino acid sequence, secondary structure and tertiary structure of 14-3-3 (Eg14-3-3) protein of Echinococcus granulosus with bioinformatics software and predict the B cell epitope and T cell epitope. Screening the dominant epitopes, the development of safe and efficient peptide vaccine and specific diagnosis of anti-lays the foundation. Methods The sequence, structure, B cell epitopes and T cell epitopes of Eg14-3-3 were predicted by DNAstar software, ProPred MHC Class-Ⅱ Binding Peptide Prediction Server and Biosun software. Results Eg14-3-3 was a protein of 247 amino acid with a relative molecular mass of 27 940. The secondary structure of Eg14-3-3 consisted mainly of 6 α-helix structures with short β-sheets and irregularly curled structures with high degree of sequence It is predicted that there are 14 B-cell epitopes and 6 T-cell epitopes in this protein. Conclusion The protein amino acid sequence, secondary structure and tertiary structure were analyzed, and 14 B-cell epitopes and 6 T-cell epitopes were predicted to exist in Eg14-3-3.
