目的:利用超高效液相色谱法测定不同批次石柑子中N-反式香豆酰酪胺的含量。方法:采用Agilent ZORBAX Eclipse XDB-C18柱(2.1mm×100mm,18μm)为色谱柱,柱温为25℃;以甲醇-水溶液为流动相,进行梯度洗脱,流速为0.2m L/min,检测波长为300nm,测定石柑子中N-反式香豆酰酪胺的含量。结果:N-反式香豆酰酪胺在进样量为6.14~73.68ng(r=0.99934)的范围内与峰面积呈良好的线性关系,所建立的含量测定方法的平均回收率为98.11%(n=9),RSD值为1.32%。结论:本研究所建立的石柑子中N-反式香豆酰酪胺的含量测定方法操作简便、准确、重复性好,可为石柑子的质量控制提供参考。
OBJECTIVE: To determine the content of N-trans-coumatron in different groups of Citrus sinensis using ultra performance liquid chromatography. Methods: The column of Agilent ZORBAX Eclipse XDB-C18 (2.1 mm × 100 mm, 18 μm) was used as the column. The column temperature was 25 ℃. The mobile phase was methanol-water. The gradient elution was carried out at a flow rate of 0.2 mL / Wavelength of 300nm, Determination of stone Citrus N-trans-coumarate tyramine content. Results: There was a good linear relationship between the peak area and the content of N-trans-coumatron in the range of 6.14-73.68 ng (r = 0.99934). The average recoveries of the established method were 98.11% (n = 9) with a RSD value of 1.32%. Conclusion: The method for the determination of N-trans-coumatron in Citrus unshiuis established in this study is simple, accurate and reproducible. It provides a reference for the quality control of Citrus unshiuilis.
