目的:探讨忍冬木层孔菌醇提物对大鼠乳腺癌癌前病变的影响,为忍冬木层孔菌防治乳腺癌提供科学依据。方法:取雌性未孕SD大鼠,制备乳腺癌癌前病变模型。连续灌胃给药4周后观察大鼠一般情况、采用放射免疫法检测大鼠血清中性激素雌二醇(E2)、孕酮(P)、催乳素(PRL)、黄体生成素(LH)和卵泡刺激素(FSH)含量,用HE染色分析乳腺组织病理变化,用免疫组化分析乳腺组织中雌激素受体α(ERα)、β(ERβ)和孕激素受体(PR)蛋白表达变化。结果:忍冬木层孔菌醇提物600mg/kg能降低血清中E2水平(P<0.05),升高P、PRL的含量(P<0.05或P<0.01),对LH和FSH未见明显的影响;150、300、600mg/kg能降低卵巢、子宫质量指数(P<0.05或P<0.01),改善乳腺组织病理变化(P<0.05或P<0.01);600mg/kg逆转乳腺组织中上调的ERα、ERβ和PR的异常表达(P<0.05或P<0.01)。结论:忍冬木层孔菌醇提物能够调节乳腺癌癌前病变大鼠体内性激素水平和效应,抑制激素异常对乳腺组织的不良刺激,从而改善大鼠乳腺组织病理变化,有效的阻止乳腺癌癌前病变的发生、发展。
OBJECTIVE: To investigate the effects of alcohol extract of Lonicera japonica on precancerous lesions of breast cancer in rats and to provide a scientific basis for the prevention and treatment of breast cancer. Methods: Female non-pregnant SD rats were used to prepare precancerous lesions of breast cancer. Four weeks after continuous intragastric administration, the general situation of rats was observed. The levels of serum estradiol (E2), progesterone (P), prolactin (PRL), luteinizing hormone (LH) Follicle stimulating hormone (FSH), the histopathological changes of breast tissue were analyzed by HE staining. The expressions of estrogen receptor alpha (ERα), beta (ERβ) and progesterone receptor (PR) in breast tissues were analyzed by immunohistochemistry. Results: The alcohol extract of Lonicera japonica 600mg / kg could reduce the level of E2 in serum (P <0.05), increase the contents of P and PRL (P <0.05 or P <0.01) (P <0.05 or P <0.01), and improve the histopathological changes of breast (P <0.05 or P <0.01). In 600 mg / kg group, the ovarian and uterine mass index Abnormal expression of ERα, ERβ and PR (P <0.05 or P <0.01). CONCLUSION: Alcohol extract of Pseudotsuga obscurus can regulate sex hormone level and effect in rats with precancerous lesions of breast, inhibit the abnormal stimulation of hormone abnormalities on mammary gland tissue, thus improve pathological changes of breast tissue in rats and effectively prevent breast cancer Pre-lesion of the occurrence and development.
