目的 探讨腺病毒载体介导的早幼粒细胞性白血病(promyelocyticleukemia ,PML)生长抑制因子抑制胆囊癌细胞系(GBC- SD)的生长作用。方法 用重组绿色荧光蛋白的腺病毒(Ad GFP)感染胆囊癌细胞系,检测其对胆囊癌细胞株的感染力及Ad- PML对细胞体外生长的影响。用DNAladdering和TUNEL法检测细胞凋亡,用流式细胞仪检测细胞周期。结果 腺病毒滴度为10 0MOI时被感染的胆囊癌细胞可达10 0 % ;PML能明显抑制GBC SD细胞体外生长(P <0 .0 5 ) ;Ad PML感染后GBC SD细胞出现DNA降解带,凋亡细胞数量明显增加(P <0 .0 1) ;但细胞周期无明显改变。结论PML能显著抑制GBC- SD细胞的体外生长,PML通过诱导胆囊癌细胞凋亡发挥其生长抑制作用。
Objective To investigate the inhibitory effect of adenovirus vector-mediated growth inhibitory factor of promyelocyticleukemia (PML) on the growth of gallbladder carcinoma cell line (GBC-SD). Methods The gallbladder carcinoma cell lines were infected with recombinant adenovirus carrying green fluorescent protein (GFP), and their effects on the infection of gallbladder carcinoma cell lines and the effect of Ad-PML on the growth of the cells were studied. Apoptosis was detected by DNAladdering and TUNEL. The cell cycle was detected by flow cytometry. Results The number of gallbladder carcinoma cells infected with adenovirus was 100% when the titer of adenovirus was 100 MOI. PML could significantly inhibit the growth of GBC SD cells in vitro (P <0.05). DNA degradation bands appeared in GBC SD cells after Ad PML infection , The number of apoptotic cells increased significantly (P <0.01), but the cell cycle did not change significantly. Conclusion PML can significantly inhibit the growth of GBC-SD cells in vitro. PML can inhibit the growth of GBC cells by inducing apoptosis.
