目的:以苦荞麦叶柄为外植体,建立一套苦荞麦植物的快速繁殖技术。方法:以MS为基本培养基,通过研究激素6-BA、2,4-D、NAA和IBA对愈伤组织诱导、不定芽分化和不定根产生的影响,找出其相应的最佳培养基配方。结果与结论:苦荞麦最佳愈伤组织诱导培养基为MS+6-BA 0.5 mg/L+2,4-D 4.0 mg/L;最佳不定芽分化培养基为MS+6-BA 2.0 mg/L+NAA 0.3 mg/L;最佳生根培养基为MS+IBA 0.5 mg/L。将组培苗移栽至松软的土壤中,注意适当遮光,组培苗成活率达90%以上。
OBJECTIVE: To establish a set of rapid propagation technology of tartary buckwheat by taking the buckwheat petiole as explant. Methods: MS-based medium was used to study the effect of hormone 6-BA, 2,4-D, NAA and IBA on the induction of callus, adventitious bud differentiation and adventitious root formation. The best medium formula . RESULTS AND CONCLUSION: The best callus induction medium for tartary buckwheat was MS + 6-BA 0.5 mg / L + 2,4-D 4.0 mg / L; the best adventitious bud differentiation medium was MS + 6-BA 2.0 mg / L + NAA 0.3 mg / L; the best rooting medium was MS + IBA 0.5 mg / L. Tissue culture seedlings transplanted to the soft soil, attention to appropriate shading, tissue culture seedlings survival rate of more than 90%.
