GAM是以gp130胞浆区为探针筛选而得到的一个新的gp130结合分子,探讨其在gp130信号传导中可能具有的重要作用的基础是证实gp130胞浆区与GAM的结合。我们采用thioredoxin融合蛋白表达系统表达了GAM分子的融合蛋白。在此基础上,发现GST-gp130融合蛋白与thioredoxin-GAM融合蛋白共沉淀,而单独GST不与thioredoxin-GAM融合蛋白共沉淀,证实了GAM与gp130的特异性结合,此结果为探讨GAM分子的功能打下了重要的基础。
GAM is a new gp130-binding molecule screened from the cytoplasmic domain of gp130 and the basis for its possible role in gp130 signaling is to confirm the binding of gp130 to GAM in the cytoplasm. We used the thioredoxin fusion protein expression system to express GAM fusion proteins. On this basis, it was found that the GST-gp130 fusion protein co-precipitated with the thioredoxin-GAM fusion protein, whereas the GST alone did not co-precipitate with the thioredoxin-GAM fusion protein, confirming the specific binding between GAM and gp130. Function laid an important foundation.
