目的 :观察外源性IL 4对K5 6 2增殖及死亡的影响。方法 :用逆转录病毒载体将IL 4cDNA导入K5 6 2细胞 ,检测其增殖反应 ,用流式细胞仪及DNA片段凝胶电泳检测细胞增殖周期及凋亡情况 ,用台盼蓝排除法计数死亡细胞。结果 :高表达IL 4的K5 6 2细胞增殖反应明显低于低表达株、亲代K5 6 2株 ;前者培养过程中死亡细胞百分率明显升高且可被IL 4单克隆抗体阻断 ,但其自发凋亡细胞数及增殖周期均无明显改变。结论 :外源性IL 4基因表达产物可诱导K5 6 2细胞死亡 ,但并非凋亡机制介导
Objective: To observe the effect of exogenous IL 4 on the proliferation and death of K562 cells. Methods: IL-4 cDNA was transduced into K562 cells by retroviral vector and the proliferative response was detected. Cell proliferation cycle and apoptosis were detected by flow cytometry and DNA fragment gel electrophoresis. The number of dead cells was counted by trypan blue exclusion . Results: The proliferation of K562 cells with high expression of IL-4 was significantly lower than that of the low-expression K562 cells. The percentage of dead cells in the former was significantly increased and could be blocked by IL-4 monoclonal antibody. However, There was no significant change in the number of apoptotic cells and the proliferation cycle. Conclusion: The exogenous IL 4 gene product can induce the death of K5 6 2 cells but is not mediated by apoptosis
