为了研究心肌肌凝蛋白轻链-1(CMLC-1)组织特异性的特点,并对其构型在心肌发育过程中的表达规律以及不同哺乳动物CMLC-1构型的进化规律进行探讨,应用抗人心室肌CMLC-1单克隆抗体(McAB)对成人和不同胎龄胎儿的心肌和其它组织、不同种属哺乳动物的心肌进行了免疫组织化学研究。结果发现:成人心室肌,慢性骨骼肌均发生强烈反应,成人心房,胎儿心室肌发生交叉反应,而其它组织如快型骨骼肌、平滑肌以及肝脾等非肌组织不发生反应。胎儿心肌的交叉反应程度随胎龄增加而增强,哺乳动物心肌均发生交叉反应,且随动物进化等级的提高呈增强趋势。结果说明:人CMLC-1具有一定的组织特异性,CMLC-1的表达与心脏发育和动物进化程度相关。
In order to study the tissue-specific characteristics of cardiac myosin light chain-1 (CMLC-1), and to study its expression pattern during myocardial development and the evolutionary rules of CMLC-1 in different mammalian tissues, Anti-human ventricular myocytes CMLC-1 monoclonal antibody (McAB) on adult and different gestational age fetal heart and other tissues, different species of mammalian myocardium immunohistochemistry. The results showed that adult ventricular muscle and chronic skeletal muscle all reacted strongly, while adult human atria and fetal ventricular muscle cross-reacted. However, other tissues such as fast skeletal muscle, smooth muscle and non-muscle tissue such as liver and spleen did not react. The degree of cross-reaction of fetal myocardium increased with the increase of gestational age, mammalian myocardium cross-reaction occurred, and with the evolution of animal evolution increased. The results showed that human CMLC-1 has certain tissue specificity, and the expression of CMLC-1 is related to heart development and animal evolution.
