本文建立cAMP-PDE活性检测的放射纸层析-液闪计数法,应用该法对八种多糖药物的cAMP-PDE抑制活性进行评价。结果发现:竹节香附提取物的脂溶性总皂甙分别以10,30,70,100μg浓度进行体外抑制兔脑cAMP-PDE,其抑制率相应为31.0%.36.0%,58.7%,61.1%;水溶性皂甙的抑制率相应为36.4%(10μg),38.6%(30μg),59.45%(50μg),50%(70μg),61.3%(100μg)。另外七种天然多糖对cAMP-PDE的抑制同样见到较好的效果,其中肝素、蘑菇浸膏、猴菇菌片、紫灵芝片和灵芝精液的抑制率均远大于阳性药物-氨茶碱(22.32%),唯云芝肝泰未见效果。
In this paper, radiopharmaceutical chromatography-liquid scintillation counting method was established for the determination of cAMP-PDE activity. The cAMP-PDE inhibitory activity of eight polysaccharide drugs was evaluated by this method. The results showed that the total fat-soluble saponins extracted from C. sinensis extract inhibited the cAMP-PDE of rabbit brain in vitro at concentrations of 10, 30, 70, and 100 μg, and the inhibitory rates were 31.0%, 36.0%, 58.7%, and 61.1%, respectively. The inhibitory rates of saponin corresponded to 36.4% (10 μg), 38.6% (30 μg), 59.45% (50 μg), 50% (70 μg), 61.3% (100 μg). In addition, the inhibition of cAMP-PDE by the other seven natural polysaccharides also showed better results. Among them, the inhibitory rates of heparin, mushroom extract, mushroom mushroom slice, purple fungus slice, and Ganoderma lucidum semen were far greater than that of the positive drug aminophylline ( 22.32%), Weiyun Zhigantai has no effect.
