杜仲(Eucommia ulmoides Oliv.)为名贵中药材。为了扩大杜仲药源,作者对杜仲愈伤组织(包括茎段、幼叶、子房、花药愈伤组织)和杜仲树皮,新鲜叶片的有效成分进行了初步研究,证明它们有一些相同的成分用2,6-二氯酚靛酚滴定法测定叶子和愈伤组织中维生素C的含量;用紫外分光光度法测定了树皮、叶片、愈伤组织中桃叶珊瑚甙的含量;用薄层层析-分光光度法测定了氯原酸含量。结果说明,杜仲皮、叶及愈伤组织中都含有维生素C、桃叶珊瑚甙、氯原酸等成分。但含量差异较大。本实验为我们利用植物组织培养这一新兴生物技术扩大杜仲药源、为研究有效成分在杜仲植物体中的形成过程提供了参考依据。
Eucommia ulmoides Oliv. is a valuable Chinese herbal medicine. In order to expand the source of eucommia, the authors conducted preliminary studies on the active ingredients of Eucommia callus tissue (including stem segments, young leaves, ovary, and anther callus) and Eucommia bark and fresh leaves, and they showed that they had some of the same components. The contents of vitamin C in leaves and callus were determined by the 2,6-dichlorophenol phenol titration method; the contents of the coral leafhoppers in the bark, leaves and callus were determined by ultraviolet spectrophotometry; Chlorogenic acid content was determined by chromatography-spectrophotometry. The results showed that the bark, leaves, and callus of Eucommia ulmoides contain vitamin C, phyllophylline, and chlorogenic acid. However, the content varies greatly. This experiment provided a reference for us to expand the source of eucommia ulmoides by using this new biological technology of plant tissue culture and to study the formation process of active ingredients in Eucommia plant.
