目的建立一种同时分离和测定大花罗布麻叶中6种黄酮类成分的微乳液相色谱方法。方法用HPLC法测定,色谱柱为Zorbax SB-C_(18)(4.6mm×150mm,5μm);流速:0.8ml·min~(-1);检测波长:360nm;柱温:30℃;采用经过优化后的微乳体系为流动相,其组成为:2.0%Genapol X-080-3.0%正丁醇-0.6%乙酸乙酯-0.2%三乙胺-94.2%水溶液(20mmol·L~(-1) H_3PO_4调节至pH 5.0)。结果大花罗布麻叶中6种黄酮类成分在15min内达到基线分离,在1~500mg·L~(-1)范围内,6个黄酮成分的线性相关系数≥0.9998,回收率98.9%~100.8%。结论本微乳液相色谱法可应用于大花罗布麻叶中6种黄酮成分的同时分离和测定。
Objective To establish a microemulsion method for the simultaneous separation and determination of six flavonoids in Apocynum leaves. The method was determined by HPLC. The chromatographic column was Zorbax SB-C 18 (4.6 mm × 150 mm, 5 μm). The flow rate was 0.8 ml · min -1. The detection wavelength was 360 nm. The column temperature was 30 ℃. The optimized microemulsion system consisted of 2.0% Genapol X-080-3.0% n-butanol -0.6% ethyl acetate-0.2% triethylamine-94.2% water solution (20 mmol·L -1 ) H 3 PO 4 to pH 5.0). Results Six flavonoids of Apocynum venetum were baseline isolated within 15 min. The linear correlation coefficients of six flavonoids were within the range of 1 ~ 500 mg · L -1, the recoveries were 98.9% -100.8 %. Conclusion This microemulsion method can be applied to the simultaneous separation and determination of six flavonoids in Apocynum leaves.
