目的建立紫草酸血药浓度的高效液相色谱测定方法,研究其在大鼠体内药代动学变化规律。方法大鼠静脉注射紫草酸10mg/kg后,于不同时间点采血,用HPLC测定其血药浓度,并用DAZ软件拟合,计算药代动力学各参数。结果紫草酸血药浓度的回归方程的线性关系良好,最低检测浓度为0.2μg/ml。药时曲线符合二室开放模型。结论首次建立了紫草酸血药浓度的HPLC测定方法。方法操作简单、专属性强、灵敏度高。静脉注射紫草酸,其体内过程的药时曲线符合二室开放模型,具有体内分布快、血药浓度下降迅速等特点。
Objective To establish a HPLC method for the determination of the concentration of lithosperminic acid in rats and study its pharmacokinetics in rats. Methods After intravenous injection of lithospermic acid (10mg / kg), rats were sacrificed at different time points to determine the plasma concentration by HPLC. The pharmacokinetic parameters were calculated by DAZ software. Results The regression equation of the concentration of lithiolinic acid had a good linear relationship. The lowest detection concentration was 0.2μg / ml. Drug curve in line with two-room open model. Conclusion For the first time, HPLC method was established for determining the concentration of lithospermic acid. The method is simple, highly specific and sensitive. Intravenous injection of purple oxalic acid, its in vivo process of drug-time curve in line with the two-compartment open model, with rapid distribution in vivo, rapid decline in plasma concentration and other characteristics.
