目的:探索运动对2型糖尿病胰岛素抵抗大鼠脂肪组织、肝脏RBP4基因mRNA表达、胰岛素敏感性的影响。方法:通过高糖高脂肪食物喂养结合小剂量链脲佐菌素部分破坏胰腺的方法建立2型糖尿病大鼠模型,随机分为对照组(A组)和游泳运动组(B组),进行8周游泳运动。健康雄性SD大鼠作为健康安静对照组(C组)。采用逆转录-聚合酶链反应(RT-PCR)法、放射免疫法观察大鼠肝脏、附睾脂肪组织RBP4mRNA表达、胰岛素敏感性的变化。结果:1)与C组相比,A组附睾脂肪组织、肝组织中RBP4基因mRNA的表达明显上调,体质量、附睾脂肪组织质量、肾周脂肪组织质量、脂体比、空腹胰岛素、胰岛素抵抗指数IRI明显增高。2)与A组相比,B组附睾脂肪组织RBP4mRNA的表达明显下调,RBP4mRNA表达与IRI呈正相关。体质量、附睾脂肪组织质量、肾周脂肪组织质量、脂体比、空腹胰岛素I、RI明显降低,肝组织中RBP4mRNA的表达无明显改变。结论:运动能够逆转2型糖尿病胰岛素抵抗鼠脂附睾肪组织RBP4mRNA表达的上调,改善胰岛素敏感性。
Objective: To explore the effect of exercise on the mRNA and protein expression of RBP4 in adipose tissue and liver of type 2 diabetic rats with insulin resistance. Methods: A rat model of type 2 diabetes mellitus (T2DM) was established by feeding high-fat and high-fat diet combined with low-dose streptozotocin to partially destroy the pancreas. The rats were randomly divided into control group (A group) and swimming group (B group) Week swimming exercise. Healthy male SD rats served as healthy and quiet control group (C group). Reverse transcription polymerase chain reaction (RT-PCR) and radioimmunoassay were used to observe the changes of RBP4mRNA and insulin sensitivity in rat liver and epididymis adipose tissue. Compared with group C, the mRNA expression of RBP4 in epididymal adipose tissue and liver tissue in group A was significantly up-regulated. The body weight, the quality of epididymal adipose tissue, the quality of peritoneal adipose tissue, fat ratio, fasting insulin and insulin resistance Index IRI increased significantly. 2) Compared with group A, the expression of RBP4 mRNA in epididymal adipose tissue of group B was significantly decreased, and the expression of RBP4 mRNA was positively correlated with IRI. Body weight, epididymal fat tissue mass, perirenal adipose tissue mass, fat ratio, fasting insulin I and RI were significantly decreased, and the expression of RBP4 mRNA in liver tissue did not change significantly. Conclusion: Exercise can reverse the up-regulation of RBP4 mRNA expression in adipose-derived adrenal tissue in type 2 diabetic rats and improve insulin sensitivity.
