目的:观察复方土茯苓颗粒对高尿酸血症大鼠血尿酸(UA),黄嘌呤氢化酶(XO)活性及肝脏中黄嘌呤氧化酶(XDH)mRNA表达水平的抑制作用。方法:48只SD大鼠随机分成正常组,模型组,非布司他组(4 mg·kg~(-1)),复方土茯苓颗粒低、中、高剂量组(1,2,4 g·kg~(-1)),除正常组外,其余各组制备高尿酸血症模型及给予相应药物干预,每天1次,共给药5 d。实验结束后测定各组大鼠的血UA,肝XO活性以及肝中的XDH mRNA表达水平。结果:模型组大鼠的血UA,肝XO活性,肝XDH mRNA表达水平均比正常组明显升高(P<0.05);经药物治疗后,各给药组大鼠血UA,肝XO活性,肝XDH mRNA表达水平均比模型组明显降低(P<0.05),复方土茯苓颗粒低剂量组与模型组无明显差别。结论:复方土茯苓颗粒可以有效降低高尿酸血症大鼠血UA,并可以抑制XO活性与肝脏中XDH mRNA表达水平。
Objective: To observe the inhibitory effect of Fufang Tufuling granule on the blood uric acid (UA), xanthine hydrolase (XO) activity and xanthine oxidase (XDH) mRNA expression in hyperuricemic rats. Methods: Forty eight Sprague-Dawley rats were randomly divided into four groups: normal group, model group, febuxostat group (4 mg · kg -1) · Kg ~ (-1)). Except for the normal group, the other groups were given hyperuricemia model and given the corresponding drug intervention once a day for 5 days. After the experiment, blood UA, liver XO activity and XDH mRNA expression in the liver of each group were measured. Results: The levels of blood UA, liver XO and liver XDH mRNA expression in model group were significantly higher than those in normal group (P <0.05). After drug treatment, blood UA, liver XO activity, Liver XDH mRNA expression levels were significantly lower than the model group (P <0.05), compound Tufuling granules low dose group and the model group no significant difference. Conclusion: Compound Tufuling Granules can effectively reduce blood UA in hyperuricemic rats and inhibit the activity of XO and the expression of XDH mRNA in the liver.
