目的:观察何首乌含药血清对体外培养的人肝细胞活性的影响,考察何首乌入血成分是否具有肝细胞毒作用。方法:采用血清药理学方法制备何首乌含药血清及空白对照组血清,作用于体外培养的人正常肝L02细胞和肝癌HepG2细胞,MTT法检测不同浓度血清对两种细胞活力的影响;倒置相差显微镜观察各浓度血清作用后两种细胞的形态变化;测定含药血清作用于两种细48 h后,细胞培养液转氨酶活性变化。结果:何首乌含药血清对人正常肝L02细胞和肝癌HepG2细胞的活力没有明显影响。两种细胞含药血清组与空白血清组相比,细胞形态都没有发生明显改变,细胞培养液转氨酶活性(ALT、AST)也没有显著差异(P>0.05)。结论:何首乌含药血清对体外培养的两种人源肝细胞活性均没有明显影响。
Objective: To observe the effect of Radix Polygoni multiflori containing serum on the activity of human hepatocytes cultured in vitro, and to investigate whether the blood components of Polygonum multiflorum have hepatotoxic effects. Methods: The serum of Polygonum multiflorum contained in serum and the blank control group were prepared by serum pharmacology. The cells were cultured in human normal liver L02 cells and HepG2 hepatoma cells. MTT assay was used to detect the effect of different concentrations of serum on the cell viability. The morphological changes of the two kinds of cells were observed after the serum of each concentration was treated. The activity of aminotransferase in the cell culture medium was measured after the serum containing the two drugs were treated for 48 hours. Results: Polygonum multiflorum contained no significant effect on the activity of human normal L02 cells and HepG2 cells. Compared with the blank serum group, the cell morphology of the two cell-containing serum groups did not change significantly, and there was no significant difference in ALT, AST (P> 0.05). CONCLUSION: Polygonum multiflorum contained no serum on the activity of two human hepatocytes cultured in vitro.
