目的观察铁皮石斛对小鼠抗氧化能力的影响。方法将清洁级雄性昆明种小鼠75只随机分为正常对照组、模型组和铁皮石斛低、中、高剂量组,每组15只。对低、中、高剂量组按125 mg/kg体重、250 mg/kg体重、500 mg/kg体重分别给予铁皮石斛灌胃,对照组给予相同体积的生理盐水灌胃。每天1次,连续30 d。检测ROS、AngⅡ、SOD、MDA、GSH-Px及NF-κBmRNA表达。结果与对照组比较,模型组ROS、AngⅡ、MDA水平,NF-κB mRNA表达均显著升高(P<0.01),而SOD、GSH-Px活力均显著下降(P<0.01);中剂量组和高剂量组ROS、AngⅡ、MDA水平、NF-κB mRNA表达均下降或显著下降(P<0.05或P<0.01),而SOD、GSH-Px活力均升高或显著升高(P<0.05或P<0.01)。结论铁皮石斛可通过提高抗氧化能力和减少氧化产物生成纠正氧化/抗氧化失衡状态,这一作用与其剂量相关。
Objective To observe the effect of Dendrobium candidum on antioxidant capacity in mice. Methods Seventy-five male Kunming mice were randomly divided into normal control group, model group and low, medium and high dose group of Dendrobium officinale, 15 rats in each group. The low, medium and high dose groups were administered with 125 mg / kg body weight, 250 mg / kg body weight and 500 mg / kg body weight respectively. The control group was given the same volume of physiological saline. 1 day, 30 consecutive days. The levels of ROS, AngⅡ, SOD, MDA, GSH-Px and NF-κB mRNA were detected. Results Compared with the control group, the levels of ROS, AngⅡ, MDA and the expression of NF-κB mRNA in the model group were significantly increased (P <0.01), while the activities of SOD and GSH-Px were significantly decreased The levels of ROS, AngⅡ, MDA and the expression of NF-κB mRNA in high-dose group were significantly decreased (P <0.05 or P <0.01), while the activities of SOD and GSH-Px were significantly increased <0.01). Conclusion Dendrobium candidum can correct the imbalance of oxidation / antioxidation by increasing the antioxidant capacity and reducing the production of oxidation products, which is related to its dosage.
