目的:探讨治伤巴布剂对福尔马林足底炎性疼痛大鼠模型背根神经节中辣椒素受体水平的影响,研究治伤巴布剂对大鼠炎性疼痛是否有镇痛作用。方法:SD大鼠36只,随机分为空白对照组(n=12)、模型组(n=12)及治疗组(n=12);空白对照组大鼠不予任何处理,模型组左后足底注射0.1 mL5%福尔马林,治疗组大鼠造模前1 d于左后足外敷治伤巴布剂,然后左后足底注射0.1 mL5%福尔马林,再一直外敷治伤巴布剂,观察造模后大鼠疼痛行为学反应,并以Dubuisson评分方法进行记录比较。各组大鼠在指定时间点取出L3-6节段背根神经节,运用实时荧光定量(RT-PCR)、蛋白质印迹(Western blot)检测辣椒素受体在大鼠背根神经节中的表达水平。结果:模型组及治疗组呈典型的双相伤害性行为反应,但治疗组大鼠疼痛反应积分低于模型组,差异有统计学意义(P<0.05﹚;实时荧光定量检查结果显示模型组辣椒素受体mRNA相对表达量大于治疗组,治疗组辣椒素受体mRNA相对表达量大于空白对照组,3组比较,差异有统计学意义(P<0.05﹚,3组组内3个时相点的相对表达量差异均无统计学意义(P>0.05);蛋白质印迹法灰度扫描显示模型组辣椒素受体相对表达量大于治疗组,治疗组辣椒素受体相对表达量大于空白对照组,3组比较,差异有统计学意义(P<0.05﹚,3组组内3个时相点的相对表达量差异均无统计学意义(P>0.05)。结论:治伤巴布剂能缓解大鼠炎性疼痛反应,对大鼠炎性疼痛具有一定的镇痛作用,可降低福尔马林足底炎性疼痛大鼠模型背根神经节中辣椒素受体的表达水平。
OBJECTIVE: To investigate the effect of cataplasm on the level of capsaicin receptor in the dorsal root ganglion of formalin-induced rat model of plantar inflammatory pain and to find out whether there is analgesic effect of cataplasm on inflammatory pain in rats effect. Methods: Thirty-six SD rats were randomly divided into blank control group (n = 12), model group (n = 12) and treatment group (n = 12) The rats were injected 0.1 mL 5% formalin into the soles of the feet. The rats in the treatment group were treated with catgut embedding on the left hind foot one day before modeling, and then 0.1 mL of 5% formalin was injected into the left posterior plantar. Analgesic behavior of rats after modeling, and recorded by Dubuisson score comparison. The rats in each group were taken out L3-6 dorsal root ganglion at designated time points. The expression of capsaicin receptor in rat dorsal root ganglion was detected by real-time fluorescence quantitative (RT-PCR) and Western blot Level. Results: The model group and the treatment group showed a typical biphasic nociceptive response, but the scores of pain response in the treatment group were lower than those in the model group (P <0.05). The real-time fluorescence quantitative results showed that the model group Compared with the treatment group, the relative expression level of angiotensin receptor mRNA was higher in the treatment group than in the blank control group (P <0.05). The three time points (P> 0.05). Western blotting showed that the relative expression of capsaicin receptor in the model group was greater than that in the treatment group, and the relative expression of capsaicin receptor in the treatment group was larger than that in the blank control group (P <0.05). There was no significant difference in the relative expression of three time points between the three groups (P> 0.05). Conclusion: The cataplasm can relieve the inflammatory pain reaction in rats, and has some analgesic effects on inflammatory pain in rats, and can reduce the dose of capsaicin in the dorsal root ganglion Receptor expression levels.
