目的研究全反式维甲酸(ATRA)对表皮鳞癌细胞株A431细胞凋亡的影响,并观察干细胞更新因子BMI-1在鳞癌A431细胞系不同分化时期的表达变化,以明确BMI-1在皮肤鳞癌细胞增殖分化中的功能。方法体外培养A431细胞,分别于ATRA作用即刻、24 h及48 h收集细胞,流式细胞仪检测不同状态下细胞凋亡变化,west-ern blot法检测不同时期BMI-1蛋白表达差异。结果随ATRA作用时间的延长可以显著提高A431细胞的凋亡率。同时24 h及48 h BMI-1表达逐渐减少,与即刻组之间差异有统计学意义(P<0.05)。结论ATRA可抑制鳞癌细胞A431增殖,并影响BMI-1在肿瘤细胞不同增殖状态的表达,同时说明干细胞更新因子BMI-1在维持皮肤鳞状细胞癌增殖中可能起关键作用。
Objective To investigate the effect of all-trans retinoic acid (ATRA) on the apoptosis of squamous cell carcinoma cell line A431 and to observe the expression of stem cell factor BMI-1 in different differentiation stages of squamous cell carcinoma A431 cell line. Function of skin squamous cell carcinoma in proliferation and differentiation. Methods A431 cells were cultured in vitro. The cells were harvested immediately after ATRA treatment for 24 h and 48 h respectively. The apoptosis of cells was detected by flow cytometry. The expression of BMI-1 protein was detected by west-ern blot assay. Results With the prolongation of ATRA treatment, the apoptosis rate of A431 cells was significantly increased. At the same time, the expression of BMI-1 decreased gradually at 24 h and 48 h, which was significantly different from the immediate group (P <0.05). CONCLUSION: ATRA can inhibit the proliferation of squamous cell carcinoma A431 and affect the expression of BMI-1 in different proliferative states of tumor cells. It also shows that stem cell factor BMI-1 may play a key role in maintaining the proliferation of squamous cell carcinoma of the skin.
