目的观察氯化镨(Pr Cl3)对小鼠骨髓细胞DNA的损伤,以及亚硒酸钠(Na_2SeO_3)的干预作用。方法试验动物为5~6周龄昆明种小鼠;试验设置12个组,即5个Pr Cl_3剂量组(7.5、15、30、60和120 mg/kg·bw)、5个Na_2SeO_3干预组(在5个PrCl3剂量的基础上分别加入0.05 mg/kg·bw的Na_2SeO_3)、2个对照组[阴性组为0.85%生理盐水,阳性组为叠氮化钠(Na N3)20 mg/kg·bw]。各组小鼠采用腹腔注射染毒,运用单细胞凝胶电泳技术研究各组小鼠DNA受损情况,并比较Na_2SeO_3干预组对小鼠骨髓细胞DNA的影响。结果在7.5~120 mg/kg·bw的PrCl_3剂量内,随着PrCl_3处理剂量的增加,头长先增加后减少,尾长(TL)、全长(CL)、尾部DNA百分含量(TDNA)、尾矩(TM)、Olive尾矩(OTM)和拖尾率(PTC)逐渐增加,且存在明显的剂量-效应关系(r_(TL)=0.95,r_(CL)=0.88,r_(TDNA)=0.99,r_(TM)=0.99,r_(OTM)=0.97,r_(PTC)=0.95);将Na_2SeO_3干预组与PrCl_3剂量组两两对应相比较,Na_2SeO_3干预组的小鼠骨髓细胞尾长、全长、尾部DNA百分含量、尾矩、Olive尾矩和拖尾率均降低,经t检验显示,除7.5和15 mg/kg·bw组尾部DNA百分含量差异无统计学意义以外,其余各组各指标差异均有统计学意义(P<0.05或P<0.01)。结论在实验浓度范围内PrCl_3致小鼠骨髓细胞核DNA损伤逐渐增强,具有遗传毒作用;加入Na_2SeO_3能够降低小鼠骨髓细胞核DNA的损伤,具有明显的干预作用。
Objective To observe the DNA damage induced by Pr Cl3 in mouse bone marrow cells and the intervention of sodium selenite (Na_2SeO_3). Methods The experimental animals were Kunming mice aged 5 to 6 weeks. Twelve groups were randomly divided into five groups: PrCl_3 (7.5,15,30,60 and 120 mg / kg · bw) and five Na_2SeO_3 groups 0.05 mg / kg · bw Na_2SeO_3 was added on the basis of 5 doses of PrCl 3 respectively), two control groups (0.85% saline in negative group and 20 mg / kg · bw sodium azide) ]. The mice in each group were injected intraperitoneally with the drug. The single cell gel electrophoresis was used to study the DNA damage in each group. The effects of Na_2SeO_3 intervention group on the DNA of mouse bone marrow cells were compared. Results Proline prolonged and then decreased with increase of PrCl_3 dose in the range of 7.5 ~ 120 mg / kg · bw of PrCl_3. TL, TD, TDNA, (TM), OTM and PTC increased gradually with significant dose-effect relationship (r TL = 0.95, r CL = 0.88, r TDNA, = 0.99, r_ (OTM) = 0.97, r_ (PTC) = 0.95) .Compared with PrCl_3 group and Na_2SeO_3 group, the tail length, Tail length, tail DNA content, tail moment, Olive tail moment and tailing rate all decreased. The t test showed that there was no significant difference in tail DNA percentage except 7.5 and 15 mg / kg · bw The indexes of each group had significant differences (P <0.05 or P <0.01). Conclusions DNA damage of PrCl_3-induced mouse bone marrow cells gradually increases with genotoxicity in the experimental concentration range. Addition of Na_2SeO_3 can reduce the DNA damage of bone marrow cells in mice, which has a significant intervention.
