目的 探讨端粒酶逆转录酶 (hTERT)和P5 3基因与甲状腺癌的发生、发展及预后的关系以及它们的相互关系。方法 应用免疫组织化学SP法检测 6 0例甲状腺癌、10例正常甲状腺组织、10例结节性甲状腺肿及 10例甲状腺腺瘤组织中hTERT和P5 3的表达。结果 hTERT和P5 3阳性率在甲状腺癌中增高 (P <0 .0 5 ) ;hTERT与P5 3阳性率在未分化癌、有淋巴结转移或Ⅲ、Ⅳ期病例中增高 (P <0 .0 5 ) ;hTERT表达与P5 3表达具有相关性 (P <0 .0 5 )。结论 hTERT与P5 3的过度表达在甲状腺癌的发生发展过程中具有重要作用 ,P5 3突变在转录水平上调控hTERT基因的表达 ,激活端粒酶可能是甲状腺癌发生发展的重要阶段 ;hTERT和P5 3在甲状腺癌中表达的变化可用于评估其生物学行为和判断预后。
Objective To investigate the relationship between telomerase reverse transcriptase (hTERT) and P53 gene and the occurrence, development and prognosis of thyroid cancer and their relationship. Methods Immunohistochemical SP method was used to detect the expression of hTERT and P53 in 60 thyroid carcinomas, 10 normal thyroid tissues, 10 nodular goiter and 10 thyroid adenoma tissues. Results The positive rates of hTERT and P53 were higher in thyroid cancer (P <0.05). The positive rates of hTERT and P53 were higher in patients with undifferentiated carcinoma, lymph node metastasis or stage Ⅲ and Ⅳ (P <0.05) ). The expression of hTERT was correlated with the expression of P53 (P <0.05). Conclusion The overexpression of hTERT and P53 play an important role in the carcinogenesis and progression of thyroid carcinoma. The mutation of P5 3 regulates the expression of hTERT gene at the transcriptional level. Activation of telomerase may play an important role in the development of thyroid carcinoma. The expression of hTERT and P5 3 in thyroid cancer expression changes can be used to assess its biological behavior and prognosis.
