采用超临界二氧化碳流体技术萃取枇杷叶中熊果酸进行了较为系统的研究。选择萃取压力、萃取温度、萃取时间及分离温度4个影响因素,运用多因素多水平法安排试验,以高效液相色谱测定熊果酸的含量。选择萃取物中熊果酸含量为试验指标,用自主提出的多因素多水平试验可视化分析方法对多维空间试验数据进行分析。找出最佳工艺范围为:萃取压力为22~30MPa,分离温度为33~37℃,萃取时间为75~80min,萃取温度影响较为复杂,可以在20~38℃内进一步试验选优确定。研究证明多因素多水平可视化分析法对于多维空间多水平试验结果分析正确,应用成功。
The extraction of ursolic acid from loquat leaves by supercritical carbon dioxide fluid technique was systematically studied. Four influencing factors, extraction pressure, extraction temperature, extraction time and separation temperature were selected. The multi-factor and multi-level method was used to arrange the test, and the content of ursolic acid was determined by HPLC. The content of ursolic acid in the extract was chosen as the experimental index, and the multidimensional spatial experiment data were analyzed by using the autonomic multi-factor and multi-level visual analysis method. The optimal process conditions were as follows: the extraction pressure was 22 ~ 30MPa, the separation temperature was 33 ~ 37 ℃, the extraction time was 75 ~ 80min, the extraction temperature had a more complicated influence and could be further optimized at 20 ~ 38 ℃. The research proves that multi-factor and multi-level visual analysis method is correct for the multi-level multi-dimensional test results and is applied successfully.
