目的 :比较永生化下颌髁突软骨细胞 (IMCC)和原代培养的下颌髁突软骨细胞 (MCC)在形态学特征等方面的差别。方法 :通过相差显微镜、光镜、电镜等观察细胞的形态特征 ,采用HPIAS_10 0 0图像分析系统比较IMCC和MCC的大小。结果 :MCC表现出以多角形为主的多形性形态特征 ,并随培养代数的增加细胞形态发生改变 ;IMCC为多角形 ,传代对其形态无明显影响。MCC的超微结构表现出软骨细胞的一般特征 ,IMCC近似于前成软骨细胞 ,部分细胞核浆比例、细胞核形态发生改变。图像分析发现IMCC在细胞长短径、面积等方面小于MCC(P <0 .0 1)。结论 :IMCC基本保留了MCC的形态特征 ,其表型稳定 ,属于一种分化程度较低、幼稚的软骨细胞 ,可能近似于下颌髁突的增殖层细胞。
OBJECTIVE: To compare morphological characteristics of mandibular condylar chondrocytes (MCCs) with immortalized mandibular condylar chondrocytes (IMCCs) and primary cultured chondrocytes. Methods: Morphological characteristics of cells were observed by phase contrast microscope, light microscope and electron microscope. The size of IMCC and MCC were compared by HPIAS_10 0 image analysis system. Results: MCC showed pleomorphic polygamorphic morphological features, and cell morphology changed with the increase of culture algebra. The IMCC was polygonal with no significant effect on its morphology. The ultrastructure of MCC showed the general characteristics of chondrocytes. IMCC was similar to pre-chondrogenic cells, with some nuclear cytoplasm proportion and nuclear morphological changes. Image analysis showed that IMCC was smaller than MCC in terms of the length, the diameter, the area, etc. (P <0.01). CONCLUSION: IMCC basically retains the morphological characteristics of MCC. Its phenotype is stable and belongs to a kind of low-grade, naive chondrocytes, which may be similar to proliferating layer cells of mandibular condyle.
