目的:初步探讨CD4+T细胞表达的腺苷CD39和CD73在过敏性哮喘患者外周血中的水平变化及与叉头蛋白3(FoxP3)、GATA结合蛋白3(GATA3)和维A酸相关孤儿受体(ROR)-γt mRNA相关性。方法:选择54例对屋尘螨过敏的慢性持续期哮喘患者[间歇状态和轻度持续哮喘组(间歇-轻度组)29例,中重度组25例]和45名健康志愿者,通过荧光实时定量PCR方法检测并比较研究对象外周血CD4+T细胞中CD39、CD73、FoxP3、GATA3和ROR-γt mRNA水平。结果:CD39 mRNA水平正常对照组明显高于间歇-轻度组(P=0.000),间歇-轻度组明显高于中重度组(P=0.001)。CD73 mRNA水平正常对照组明显高于间歇-轻度组(P=0.001),哮喘亚组间差异无统计学意义(P=0.126)。哮喘组CD4+T细胞表达的CD39 mRNA与FoxP3 mRNA水平呈显著正相关性(r=0.607,P=0.000),与GATA3 mRNA水平呈显著负相关性(r=-0.505,P=0.000),与ROR-γt mRNA无显著相关性(r=-0.287,P=0.054)。哮喘组CD4+T细胞表达的CD73 mRNA与FoxP3和ROR-γt mRNA水平无显著相关性(r=0.110,P=0.450;r=-0.227,P=0.138),与GATA3mRNA水平呈显著负相关性(r=-0.286,P=0.040)。结论:CD4+T细胞表达的CD39和CD73在过敏性哮喘免疫反应过程中起到重要作用。
OBJECTIVE: To investigate the changes of peripheral blood levels of CD4 + T cells expressing adenosine CD39 and CD73 in patients with allergic asthma and the correlation between FoxP3, GATA3 and orpigan-related orphan Body (ROR) -γt mRNA correlation. Methods: A total of 54 patients with chronic persistent asthma who were allergic to house dust mite (29 in intermittent and mild persistent asthma group and 25 in moderate and severe group) and 45 healthy volunteers were enrolled in this study. Fluorescence The levels of CD39, CD73, FoxP3, GATA3 and ROR-γt mRNA in CD4 + T cells from peripheral blood were detected by real-time PCR. Results: The level of CD39 mRNA in normal control group was significantly higher than that in intermittent - mild group (P = 0.000), intermittent - mild group was significantly higher than that in moderate - severe group (P = 0.001). The level of CD73 mRNA in normal control group was significantly higher than that in intermittent - mild group (P = 0.001). There was no significant difference between subgroups of asthma (P = 0.126). There was a significant positive correlation between the expression of CD39 mRNA and FoxP3 mRNA in asthmatic CD4 + T cells (r = 0.607, P = 0.000) and negatively correlated with GATA3 mRNA (r = -0.505, P = 0.000) ROR-γt mRNA had no significant correlation (r = -0.287, P = 0.054). The expression of CD73 mRNA in CD4 + T cells in asthmatic group was not significantly correlated with FoxP3 and ROR-γt mRNA levels (r = 0.110, P = 0.450; r = -0.227, P = 0.138) r = -0.286, P = 0.040). Conclusion: CD39 and CD73 expressed by CD4 + T cells play an important role in the immune response to allergic asthma.
