目的:检测细胞增殖相关基因人特异性蛋白激酶-15(specific protein kinase 15,NYD-SP15)基因在增生牙龈及正常牙龈组织中的表达,初步探讨该基因与牙龈增生发生的关系。方法:应用RT-PCR及免疫组化法检测27例正常牙龈组织(取自外伤冠折后行牙冠延长术中切除的牙龈组织)及40例牙龈增生患者(以增生表现为主的慢性牙龈炎12例、药物性牙龈增生13例、牙龈瘤15例)牙龈切除术中切除的增生牙龈组织标本中NYD-SP15 mRNA及蛋白的表达。结果:增生牙龈组织中NYD-SP15的mRNA水平显著高于正常牙龈组织(P<0.05)。免疫组化结果显示,在正常牙龈组织中该基因高表达于胞浆,在增生牙龈组织中高表达于胞核。结论:NYD-SP15表达量异常以及定位差异在人牙龈增生发生发展过程中起到一定的作用,但该基因高表达与牙龈增生发生的相关机制尚待进一步研究。
OBJECTIVE: To detect the expression of human specific protein kinase 15 (NYD-SP15) gene in proliferating gingival and normal gingival tissues and to explore the relationship between the gene and gingival hyperplasia. Methods: Twenty-seven normal gingival tissues (gingival tissues removed from the crown during crown extension) and 40 gingival hyperplasia patients (mainly chronic gingival hyperplasia) were examined by RT-PCR and immunohistochemistry Inflammation in 12 cases, drug-induced gingival hyperplasia in 13 cases and gingival tumor in 15 cases). The expression of NYD-SP15 mRNA and protein in the gingival tissue specimens was examined. Results: The mRNA level of NYD-SP15 in proliferating gingival tissues was significantly higher than that in normal gingival tissues (P <0.05). Immunohistochemistry showed that the gene was highly expressed in the cytoplasm in normal gingival tissues and highly expressed in the gingival tissue in the hyperplasia. CONCLUSION: The abnormal expression of NYD-SP15 and the difference in localization play a role in the development of human gingival hyperplasia. However, the relationship between the high expression of NYD-SP15 and the mechanism of gingival hyperplasia remains to be further studied.
