增殖细胞核抗原(PCNA)是判断肿瘤增殖分化程度的指标之一。PCNA的免疫反应性常受到某些因素的影响,其中组织标本的固定时间对其影响最大。为此,我们观察了固定时间对PCNA表达的影响。采用LASB法检测了4例肝癌、4例胃腺癌和4例直肠癌病人,标本置于10%中性缓冲液福尔马林中固定1.5,3.0,6.0,12.0,24.0h和30.0h后,检测PCNA阳性细胞数。结果显示,随着固定时间的延长,PCNA阳性细胞逐渐减少。
Proliferating cell nuclear antigen (PCNA) is one of the indicators to judge the degree of tumor proliferation and differentiation. The immune response of PCNA is often affected by certain factors, of which the fixed time of the tissue specimen has the greatest impact on it. To this end, we observed the effect of fixed time on PCNA expression. Four patients with liver cancer, 4 patients with gastric adenocarcinoma, and 4 patients with rectal cancer were examined by LASB. Specimens were fixed in formalin with 10% neutral buffer. 1.5, 3.0, 6.0, 12.0, 24 After .0 h and 30.0 h, the number of PCNA-positive cells was measured. The results showed that as the fixation time prolonged, PCNA-positive cells gradually decreased.
