目的研究乌苏瑞宁(verussurinine,VSRN)对家兔血液流变学和血凝时间参数及小鼠出血、凝血时间的影响,以查明乌苏瑞宁是否为乌苏里藜芦总碱的抗栓活性成分。方法应用血液流变仪测定低切和高切全血比黏度、血浆比黏度;应用血凝仪测定凝血酶时间(TT)和复钙时间(RT);应用鼠尾横切法和毛细玻璃管法分别测定出血、凝血时间。结果家兔和小鼠iv4种不同剂量的乌苏瑞宁导致家兔全血比黏度及血浆比黏度呈剂量依赖性降低;TT和RT显著延长;小鼠出血、凝血时间明显延长。结论乌苏瑞宁能够显著降低家兔血液黏度,延长家兔TT和RT以及小鼠出血、凝血时间。
Objective To investigate the effect of verussurinine (VSRN) on hemorheology and hemagglutination time parameters of rabbits and the bleeding and clotting time of mice to ascertain whether wusuanruining is an anti-thrombotic active ingredient of total alkaloids from Usutra. Methods Blood rheometer was used to measure the specific viscosity of low and high cut whole blood and plasma specific viscosity. Thrombin time (TT) and recalcification time (RT) were measured with a blood coagulation meter; Cross-cutting method and capillary glass tube were applied. Methods were used to determine bleeding and coagulation time. RESULTS: Four different doses of wusu raynin in rabbits and mice resulted in a dose-dependent decrease in the viscosity and specific viscosity of plasma in rabbits; TT and RT significantly prolonged; the bleeding and coagulation time of mice were significantly prolonged. Conclusion Ussurinin can significantly reduce the blood viscosity of rabbits, prolong TT and RT in rabbits, and the time of bleeding and clotting in mice.
