目的:探讨缺氧诱导因子α(HIF-1α)、葡萄糖转运蛋白-1(GLUT-1)的表达与宫颈癌血管生成的关系。方法:应用免疫组织化学方法检测宫颈鳞状细胞癌54例中HIF-1α、GLUT-1的表达,利用CD105标记肿瘤微血管密度。结果:HIF-1α在宫颈鳞状细胞癌中的阳性表达率为50%,表达水平与分化程度、浸润深度明显相关(r=-0.674,P<0.05和r=0.897,P<0.001),与临床分期、肿瘤体积、淋巴结转移无明显相关(P>0.05);GLUT-1在子宫颈鳞状细胞癌中的表达率为57.1%,表达水平与肿瘤体积、临床分期明显相关(r=0.896,P<0.05和r=0.842,P<0.001),与分化程度、淋巴结转移、浸润深度之间无明显相关(P>0.05);HIF-1α表达水平与微血管密度之间存在明显相关(r=0.701,P<0.05),GLUT-1表达水平与微血管密度之间不存在明显相关(P>0.05);微血管密度与肿瘤体积、浸润深度之间存在明显相关(r=0.609,P<0.05和r=0.845,P<0.001),与临床分期、分化程度、淋巴结转移之间无明显相关(P>0.05);GLUT-1的表达水平与HIF-1α的表达水平之间存在明显相关(r=0.731,P<0.05)。结论:HIF-1α、GLUT-1都参与宫颈鳞状细胞癌缺氧诱导新生血管过程,并且与微血管密度密切相关,其中HIF-1α发挥重要作用。
Objective: To investigate the relationship between the expression of hypoxia inducible factor α (HIF-1α) and glucose transporter 1 (GLUT-1) and angiogenesis in cervical cancer. Methods: Immunohistochemistry was used to detect the expression of HIF-1α and GLUT-1 in 54 cases of cervical squamous cell carcinoma and the tumor microvessel density was marked by CD105. Results: The positive expression rate of HIF-1α in cervical squamous cell carcinoma was 50%. The expression level of HIF-1α was significantly correlated with the degree of differentiation and depth of invasion (r = -0.674, P <0.05 and r = 0.897, P <0.001) There was no significant correlation between clinical stage, tumor volume and lymph node metastasis (P> 0.05). The positive expression rate of GLUT-1 in cervical squamous cell carcinoma was 57.1%, the expression level was correlated with tumor volume and clinical stage (r = 0.896, There was no significant correlation between the expression of HIF-1α and the microvessel density (r = 0.701, P <0.05 and r = 0.842, P <0.001) , P <0.05). There was no significant correlation between the expression of GLUT-1 and the microvessel density (P> 0.05). There was a significant correlation between microvessel density and tumor volume and depth of invasion (r = 0.845, P <0.001). There was no significant correlation between the expression of GLUT-1 and the expression of HIF-1α (r = 0.731, P <0.05). Conclusion: Both HIF-1α and GLUT-1 are involved in the process of hypoxia-induced neovascularization in cervical squamous cell carcinoma and are closely related to the microvessel density. Among them, HIF-1α plays an important role.
