目的观察敷胸膏对RSV肺炎大鼠组织NF-κB p65及TLR4的表达的影响。方法 Wistar大鼠随机分为正常组、模型组、君臣组(敷胸膏中君臣药组成)、敷胸膏组,每组10只,除正常组外,均采用鼻腔接种RSV Long株法诱导幼龄大鼠肺炎,正常组用不含病毒的生理盐水滴鼻。于感染当天,君臣组和敷胸膏组分别于大鼠背部外敷进行干预。观察各组大鼠的一般状态、肺组织形态学,各组大鼠在感染7天后处死,采用免疫组化染色法检测各组大鼠肺组织中NFB p65和TLR4的表达。结果与正常组相比,模型组大鼠的肺组织中NF-κB p65、TLR4的表达显著增高(P<0.01),敷胸膏组和君臣组中NF-κB p65、TLR4的表达与模型组比较均明显降低(P<0.05),敷胸膏组作用较君臣组明显,具有显著的差异性(P<0.05)。结论敷胸膏干预RSV肺炎大鼠的作用机制可能与抑制大鼠肺组织中NF-κB p65、TLR4表达有关。
Objective To observe the effect of Fuzhi Paste on the expression of NF-κB p65 and TLR4 in RSV pneumonia rats. Methods Wistar rats were randomly divided into normal group, model group, monarch group (chest dressing cream King Jun Chen composition), chest cream group, 10 in each group, except the normal group, were vaccinated with nasal RSV Long Strain Age rats with pneumonia, the normal group with saline-free nasal drops. On the day of infection, the monarch group and the chest-dressing cream group were intervened in the back of rats respectively. The general condition of the rats in each group was observed. The morphology of the lung tissue was observed. The rats in each group were sacrificed 7 days after infection. The expressions of NFB p65 and TLR4 in the lung tissue of each group were detected by immunohistochemical staining. Results Compared with the normal group, the expression of NF-κB p65 and TLR4 in the model group was significantly increased (P <0.01). The expression of NF-κB p65 and TLR4 in the model group was significantly higher than that in the model group (P <0.05). The effect of Chestnut cream group was more obvious than that of Junchen group, with significant difference (P <0.05). Conclusion The mechanism of action of Chestnut cream on rats with RSV pneumonia may be related to the inhibition of the expression of NF-κB p65 and TLR4 in rat lung.
