目的:建立一种准确、快速高效毛细管电泳-紫外检测法测定人血清精氨酸含量的方法。方法:采用高效毛细管电泳法,石英毛细管(70 cm×75μm,有效长度58 cm),分离电压15 kv,检测波长254 nm,高压进样15 s,电泳缓冲溶液比例20 mmol/L pH9.33四硼酸钠:166 mmol/L SDS∶乙腈=10∶10∶3。结果:线性范围为0.03 mmol/L~1.50 mmol/L(r=0.998);以噪声比(S/N)=2时浓度为最低检测限为0.01 mmol/L,精密度为2.4%~4.0%,重复性为2.3%~2.9%,回收率为90.5%~114.2%;检测烧伤患者住院后血中精氨酸含量变化,14 d含量达到高峰。结论:该方法样品预处理简单,分离效果好,可用于实际样品的分析。
Objective: To establish an accurate, rapid and efficient capillary electrophoresis - UV detection method for the determination of human serum arginine content. Methods: High-performance capillary electrophoresis was used with a quartz capillary tube (70 cm × 75 μm and an effective length of 58 cm). The separation voltage was 15 kv and the detection wavelength was 254 nm. The sample was injected under high pressure for 15 s. The proportion of electrophoresis buffer solution was 20 mmol / Sodium borate: 166 mmol / L SDS: acetonitrile = 10:10: 3. Results: The linear range was 0.03 mmol / L ~ 1.50 mmol / L (r = 0.998). The detection limit was 0.01 mmol / L with the S / N ratio of 2 and the precision was 2.4% -4.0% , The repeatability was 2.3% ~ 2.9% and the recovery rate was 90.5% ~ 114.2%. The content of arginine in blood was detected after hospitalization and reached its peak on 14th day. Conclusion: This method is simple sample pretreatment, separation effect is good, can be used for the actual sample analysis.
