氟尿嘧啶(5-Fu)作为肿瘤的化疗药品已有近三十年的历史,由于5-Fu对肝和肾的损害,并且不同的病人对其吸收上的差别,所以检测血清中5-Fu的浓度,在指导5-Fu给药方面有一定的意义。 关于测定5-Fu血清浓度的报道很多,包括高效液相色谱和气相色谱法,其中所有的高效液相色谱法都需要一个较繁琐的处理或者柱前衍生化过程,才能获得较好的灵敏度,本文提出了一个简单而可行的高效液相色谱测定血清中5-Fu的方法,获得了较好的检测灵敏度、准确度和精密度,现报道如下。
Fluorouracil (5-Fu) has been used as a tumor chemotherapeutic drug for nearly thirty years. Due to the damage to liver and kidneys caused by 5-Fu and the difference in its uptake by different patients, the detection of 5-Fu in serum Concentration, in guiding the 5-Fu administration has some significance. There are many reports on the determination of 5-Fu serum concentration, including high performance liquid chromatography and gas chromatography, in which all of the high performance liquid chromatography requires a more complicated treatment or pre-column derivatization process in order to obtain better sensitivity, This paper presents a simple and feasible method for the determination of 5-Fu in serum by HPLC, obtaining better detection sensitivity, accuracy and precision, are reported as follows.
