抗癌方对C57/BL6J小鼠移植性肺癌的药效研究

目的:从动物实验角度明确医院自拟抗癌扶正协定方对肺癌的药效作用,裨益临床的治疗。方法:本实验考察了抗癌扶正协定方和与肺癌特效化疗顺铂联合作用对小鼠一般体征和生命周期的观察、对移植瘤C57BL/6小鼠体质量、肿瘤体积和重量/肺脏指数、白细胞、红细胞和肺组织的影响。结果:(1)单纯用中药组的小鼠体征未见明显异常,而醇提联合顺铂组小鼠一般体征最差,醇提物联合顺铂组对延长小鼠生命期的效果最差。(2)由于肿瘤的不断增大,模型组、醇提组小鼠体质量也呈增长趋势,但抗癌扶正协定方醇提物逐渐发挥抗癌疗效,肿瘤生长减慢,体质量增长减慢;顺铂有不良反应,小鼠体质量短期内下降较明显。醇提物与顺铂联用并未体现增效减毒功效,反而产生拮抗作用。(3)顺铂和醇提物均具有抑瘤作用,但以顺铂疗效较佳。醇提物联合顺铂治疗后醇提物联合顺铂产生明显拮抗作用。(4)由于肺脏指数=肺重量/小鼠体质量,醇提联合顺铂组小鼠肺脏指数最小,说明肺萎缩程度最严重。可见,顺铂抗癌效果最佳,但当与醇提物联用时,产生拮抗作用,加重不良反应,失去抗肿瘤疗效。(5)各组小鼠RBC数目比较,没有明显差距;对于WBC数目,顺铂组、醇提组WBC数量多,与模型组比较,有显著性差异(P<0.05);醇提物联合顺铂组小鼠WBC数量少,与模型组比较,有显著性差异(P<0.01);醇提物联合顺铂使用,白细胞数目显著降低,可见醇提物会显著增强顺铂毒性,产生较大的白细胞杀伤作用。(6)各图片直观比较结果:模型组癌细胞最多,且出现成块聚集现象,染色深,观察明显。3个治疗组相比,顺铂组癌细胞数目最少,醇提组居中,醇提取物联合顺铂组较多。癌细胞数目直观的反应顺铂具有较强的抗癌活性,顺铂与醇提物联合使用时,抗癌活性下降。结论:抗癌扶正协定方醇提取物有明显的肿瘤抑制作用,但对白细胞也有一定的影响。 OBJECTIVE: To clarify the pharmacological effects of self-designed anti-cancer and right-aide agreement on lung cancer from the perspective of animal experiments and to benefit clinical treatment. Methods: In this experiment, we observed the general signs and the life cycle of the mice treated with the combination of anti-cancer and righting-cure protocol and cisplatin combined with chemotherapy of lung cancer. The body mass, tumor volume and weight / lung index of C57BL / Leukocyte, erythrocyte and lung tissue effects. Results: (1) There was no obvious abnormality in the mice of the traditional Chinese medicine group, but the alcohol and cisplatin group had the poorest signs. (2) The body mass of mice in model group and alcohol extraction group also showed an increasing trend due to the increase of tumor. However, the alcohol extract of anticancer right-helping agreement gradually exerted anticancer efficacy, the growth of tumor slowed down and the growth of body weight slowed down ; Cisplatin adverse reactions, the body weight of mice decreased significantly in the short term. Alcohol extract and cisplatin combination did not reflect the synergistic effect of attenuating, but antagonistic effect. (3) Both cisplatin and alcohol extract have antitumor effects, but cisplatin has better curative effect. Alcohol extract combined with cisplatin alcohol extract combined with cisplatin obvious antagonism. (4) Due to the lung index = lung weight / body weight of mice, alcohol combined cisplatin group had the lowest lung index, indicating the degree of lung atrophy is the most serious. Can be seen, cisplatin anticancer effect of the best, but when combined with the alcohol extract, antagonistic effect, aggravating adverse reactions, loss of anti-tumor effect. (5) There was no significant difference in the number of RBC in each group. For WBC number, there were more WBC in cisplatin group and alcohol group compared with model group (P <0.05) Compared with the model group, the number of WBC in platinum group was small, which was significantly different from that in model group (P <0.01). Alcohol extract combined with cisplatin significantly decreased the number of white blood cells, and alcohol extract significantly enhanced the toxicity of cisplatin, Leukocyte killing effect. (6) Intuitive comparison of each picture: The model group had the largest number of cancer cells and aggregated agglomeration, and the staining was deep and the observation was obvious. Compared with the three treatment groups, the number of cancer cells in the cisplatin group was the lowest, alcohol group was the middle one, alcohol extract combined with cisplatin group was more. The number of cancer cells Intuitive response to cisplatin has strong anticancer activity, cisplatin and alcohol extract combination, the anticancer activity decreased. Conclusion: The anticancer Fuzheng prescription alcohol extracts have obvious tumor inhibition, but also have a certain impact on leukocytes.
