目的:探讨去甲斑蝥素(NCTD)对人乳腺癌细胞血管内皮生长因子(VEGF)表达的影响及作用机制。方法:应用四甲基偶氮唑盐(MTT)法检测去甲斑蝥素对人乳腺癌细胞系MDA-MB-231和MCF-7细胞增殖的影响;应用酶联免疫吸附试验(ELISA)检测VGEF蛋白的分泌水平;采用半定量逆转录-聚合酶链反应(RT-PCR)技术检测VEGF mRNA表达水平。结果:去甲斑蝥素对DA-MB-231和MCF-7细胞均有明显增殖抑制效应。去甲斑蝥素降低了VEGF分泌水平、VEGF mRNA的表达水平。并呈剂量-时间效应关系。结论:去甲斑蝥素能抑制人类乳腺癌细胞增殖和细胞VEGF mRNA的表达。
Objective: To investigate the effect of norcantharidin (NCTD) on the expression of vascular endothelial growth factor (VEGF) in human breast cancer cells and its mechanism of action. METHODS: The effect of norcantharidin on proliferation of human breast cancer cell lines MDA-MB-231 and MCF-7 cells was detected by MTT assay; VGEF was detected by enzyme-linked immunosorbent assay (ELISA). The level of protein secretion; using semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) technology to detect VEGF mRNA expression levels. RESULTS: Norcantharidin significantly inhibited the proliferation of DA-MB-231 and MCF-7 cells. Norcantharidin reduced VEGF secretion and VEGF mRNA expression. And showed a dose-time effect relationship. Conclusion: Norcantharidin can inhibit the proliferation of human breast cancer cells and the expression of cellular VEGF mRNA.
