目的观察黄连素对高脂膳食加小剂量链脲佐菌素(STZ)诱导的2型糖尿病(2-DM)大鼠胰岛素抵抗指数及肾周脂肪组织脂联素基因mRNA表达的影响。方法选用高脂饲料加小剂量STZ建立2-DM大鼠模型共40只,将2-DM大鼠随机分为模型组(水)和黄连素高、中、低剂量组(0.20、0.10、0.05g/kg),每组10只,另选10只大鼠为正常组,各组灌胃8周后,空腹取材,采用逆转录聚合酶链式反应检测肾周白色脂肪组织脂联素基因mRNA水平,放射免疫法测定空腹胰岛素并计算胰岛素抵抗指数(IRI)。结果正常对照组大鼠与2型糖尿病模型组大鼠脂联素基因表达、IRI间比较差异有统计学意义(P<0.01);模型组与黄连素高、中剂量组大鼠脂联素基因表达、IRI间差异有统计学意义(P<0.01)。结论黄连素可增加2-DM大鼠脂联素基因mRNA的表达,降低IRI。
Objective To observe the effect of berberine on insulin resistance index and mRNA expression of adiponectin gene in perirenal adipose tissue of type 2 diabetic rats (2-DM) induced by high-fat diet plus low-dose streptozotocin (STZ). Methods A total of 40 2-DM rat models were established using high-fat diet and low-dose STZ. 2-DM rats were randomly divided into model group (water) and berberine high, middle and low dose groups (0.20, 0.10, 0.05). g/kg), 10 rats in each group and 10 rats in the normal group. After 8 weeks of gavage in each group, the animals were vaccinated, and mRNA for adiponectin mRNA in perirenal white adipose tissue was measured by reverse transcription polymerase chain reaction. At the level, fasting insulin was measured by radioimmunoassay and the insulin resistance index (IRI) was calculated. Results The difference of adiponectin gene expression and IRI between normal control group and type 2 diabetic model group was statistically significant (P<0.01). The adiponectin gene was found in model group and high and middle dose group of berberine. The difference between expression and IRI was statistically significant (P<0.01). Conclusion Berberine can increase the mRNA expression of adiponectin and decrease IRI in 2-DM rats.
