目的:探讨冬虫夏草对糖尿病大鼠肾组织中骨桥蛋白(OPN)表达的影响。方法:将Wistar大鼠建成链脲佐菌素诱导的糖尿病模型。设对照组(VH组);糖尿病肾病组(DN组);冬虫夏草治疗组(CS组)。16周后观察各组大鼠血糖、血肌酐、24h尿蛋白排泄量、肾组织血管紧张素(Ang)Ⅱ含量等指标及肾间质纤维化评分,并用免疫组织化学方法和Northern blotting法分别观察肾组织OPN、单核巨噬细胞表面特异性标志抗原(ED-1)的表达及OPNmRNA的活性。结果:冬虫夏草能降低血糖、血肌酐、24h尿蛋白排泄量、肾组织AngⅡ含量,减轻肾间质纤维化,减少肾组织OPN的表达、OPNmRNA的活性;同时间质单核巨噬细胞浸润显著减少,且与肾小管骨桥蛋白的表达呈正相关。结论:冬虫夏草的肾脏保护作用可能与下调肾组织OPNmRNA活性介导的间质巨噬细胞的浸润有关。
Objective: To investigate the effect of Cordyceps sinensis on osteopontin (OPN) expression in the kidney of diabetic rats. Methods: Wistar rats were established as streptozotocin-induced diabetic model. The control group (VH group), diabetic nephropathy group (DN group) and Cordyceps sinensis treatment group (CS group). After 16 weeks, the blood glucose, serum creatinine, 24h urinary protein excretion, the content of angiotensin (Ang) Ⅱ and the interstitial fibrosis score of rats in each group were observed and observed by immunohistochemistry and Northern blotting Expression of OPN, ED-1 and OPN mRNA in renal tissues. Results: Cordyceps sinensis can reduce blood glucose, serum creatinine, 24h urinary protein excretion, Ang Ⅱ levels of renal tissue, reduce renal interstitial fibrosis, reduce OPN expression in kidney tissue, OPN mRNA activity; meanwhile interstitial monocyte-macrophage infiltration decreased significantly , And was positively correlated with the expression of osteopontin. CONCLUSION: The protective effect of Cordyceps sinensis on kidney may be related to the decrease of infiltration of interstitial macrophages mediated by OPN mRNA in renal tissues.
