目的:建立测定兔血浆曲安奈德浓度的高效液相色谱法。方法:应用Agilent1100系列高效液相色谱仪,以ZORBAX Eclipse XDBC18为色谱柱,以甲醇-水(52.5∶47.5)为流动相,流速为1.0mL·min-1,检测波长为240nm。血浆用乙醋乙酯-正己烷(4∶1)萃取2次后复溶检测,内标物为地西泮。结果:血浆曲安奈德在20~2000μg·L-1范围内线性关系良好(r=0.9998),最低定量质量浓度为20μg·L-1,平均回收率为(99.6±5.5)%,日内、日间RSD均小于8%。结论:该方法简单、准确,适合于兔血浆中曲安奈德药物浓度检测。
Objective: To establish a high performance liquid chromatographic method for the determination of triamcinolone acetonide in rabbit plasma. Methods: Agilent1100 series high performance liquid chromatography with a ZORBAX Eclipse XDBC18 column with methanol-water (52.5:47.5) as mobile phase at a flow rate of 1.0 mL · min-1 and a detection wavelength of 240 nm. The plasma was extracted with ethyl acetate-n-hexane (4: 1) twice and reconstituted. The internal standard was diazepam. Results: The linearity of triamcinolone acetonide was good (r = 0.9998) in the range of 20-2000μg · L-1 and the lowest mass concentration was 20μg · L-1. The average recovery was (99.6 ± 5.5)%, Inter-RSD is less than 8%. Conclusion: This method is simple and accurate and suitable for the determination of triamcinolone acetonide in rabbit plasma.
