目的探讨安多霖对微波辐射致大鼠脑损伤的防治作用。方法采用30 mW/cm2的微波辐射102只雄性SD大鼠,并于辐射前后给予12 g/kg/d的安多霖每日1次,连续7 d或14 d。在停药后3 d、4 d、5 d、6 d采用Morris水迷宫检测大鼠学习与记忆能力,于辐射后3 d、7 d、14 d和21 d采用光镜和电镜观察脑组织结构的变化,采用图像分析方法对突触间隙宽度、突触后致密物(posts-ynaptic density,PSD)厚度和致密物长度进行定量检测。结果30 mW/cm2微波辐射后大鼠学习记忆能力下降,长期药物治疗组与正常对照组相比无明显差异;30 mW/cm2微波辐射后大脑皮层和海马组织结构和超微结构损伤,表现为神经元固缩,核染色质浓缩、边集,血管周组织间隙增宽,线粒体肿胀、空化,突触间隙不清,穿孔,后膜致密物增厚。药物预防组大鼠脑组织损伤较辐射组为轻,而药物治疗组脑组织损伤明显较辐射组轻。结论30 mW/cm2微波辐射后可损伤大鼠学习记忆能力和脑组织结构,给予12 g/kg/d安多霖对微波辐射所致的上述损伤有一定的保护作用,且治疗性给药保护效果优于预防性给药。
Objective To investigate the preventive and therapeutic effects of Andorolin on brain injury induced by microwave in rats. Methods 102 male Sprague Dawley rats were irradiated with 30 mW / cm2 of microwave radiation, and 12 g / kg / d of Andorol was given once a day for 7 days or 14 days before and after irradiation. The learning and memory abilities of rats were detected by Morris water maze 3 days, 4 days, 5 days and 6 days after the drug withdrawal, and the brain tissue structure was observed by light and electron microscope on the 3rd, 7th, 14th and 21st day after radiation , The synaptic cleft width, the thickness of postsynaptic density (PSD) and the length of the dense substance were quantitatively detected by image analysis. Results After 30 mW / cm2 microwave irradiation, the ability of learning and memory in rats decreased. There was no significant difference between the long-term drug treatment group and the normal control group. The ultrastructure and ultrastructural changes of the cerebral cortex and hippocampus after 30 mW / cm2 microwave irradiation showed Neuronal pyknosis, nuclear chromatin condensation, edge set, perivascular tissue gap widened, mitochondria swelling, cavitation, synaptic cleft, perforation, thickening of the membrane after the dense matter. Compared with radiation group, the injury of brain tissue of rats in drug prevention group was lighter than that of radiation group, while the brain tissue injury of drug treatment group was lighter than that of radiation group. Conclusions After 30 mW / cm2 microwave radiation, the learning and memory abilities and brain tissue structure of rats can be damaged. At the dose of 12 g / kg / d, Andolin can protect the above injury induced by microwave radiation, and the therapeutic drug protection The effect is better than prophylactic.
