目的探讨苦参总碱对血管内皮细胞(ECV304)增殖和迁移的影响。方法采用MTT比色法分析测定苦参总碱对血管内皮细胞(ECV304)增殖的影响;流室系统检测生理剪切流场下(1.5Pa)血管内皮细胞(ECV304)。结果用0,0.5,0.75,1,2.5,5g·L-1苦参提取物培养ECV304细胞24h后,其增殖抑制率分别是0,6.97%,11.37%,29.27%,35.43%,39.26%。在生理剪切流场下,苦参总碱可显著抑制ECV304细胞的迁移(P<0.01)。结论苦参提取物抑制内皮细胞(ECV304)增殖和迁移。
Objective To investigate the effects of Sophora alkaloids on the proliferation and migration of vascular endothelial cells (ECV304). Methods MTT assay was used to determine the effect of matrine on the proliferation of vascular endothelial cells (ECV304). Flow cytometry was used to detect the endothelial cells (ECV304) under physiological shear flow (1.5 Pa). Results The proliferation inhibition rate of ECV304 cells cultured for 0, 0.5, 0.75, 1, 2.5 and 5 g · L-1 of Sophora flavescens extracts for 24 h were 0, 6.97%, 11.37%, 29.27%, 35.43% and 39.26%, respectively. Under the physiological shear flow field, matrine alkaloids significantly inhibited the migration of ECV304 cells (P <0.01). Conclusion Sophora extract inhibits the proliferation and migration of endothelial cells (ECV304).
