目的观察丙烯腈(ACN)对中国仓鼠肺成纤维细胞株(V79)的毒性作用及对线粒体膜电位、生物膜的完整性和超微结构的影响,以探讨ACN的膜毒性。方法以离体培养的V79细胞为研究对象,台盼蓝染色法观察不同染毒剂量的细胞毒性;电镜技术观察细胞生物膜超微结构,应用流式细胞仪和激光共聚焦显微镜检测线粒体膜电位、生物膜完整性。结果ACN染毒后,除1μmol/L组[(91.02±2.17)%]外,其余各处理组的细胞存活率与对照组的差异均有显著性(P<0.05);8、40、200、1 000、2 000、5 000μmol/L组线粒体膜电位改变率[(14.43±3.44)、(22.50±1.50)、(37.96±2.23)、(59.37±5.34)、(93.14±1.23)、(100.00±1.63)%]升高,与对照组[(7.84±0.61)%]的差异有显著性(P<0.05);另外V79细胞生物膜的完整性和超微结构亦发生变化。结论ACN能通过改变线粒体膜电位、膜通透性及超微结构诱导V79细胞生物膜损伤。
Objective To investigate the toxic effects of acrylonitrile (ACN) on Chinese hamster lung fibroblast cell line (V79) and its effect on mitochondrial membrane potential, biofilm integrity and ultrastructure in order to investigate the membrane toxicity of ACN. Methods V79 cells cultured in vitro were studied. Trypan blue staining was used to observe the cytotoxicity of different doses. The ultrastructure of the biofilm was observed by electron microscopy. The mitochondrial membrane potential was measured by flow cytometry and laser confocal microscopy Biofilm integrity. Results After ACN exposure, the cell survival rates of all treatment groups were significantly different from those of the control group (P <0.05) except 1 μmol / L group [(91.02 ± 2.17)%]; 8, 40, The change rates of mitochondrial membrane potential in 1 000, 2 000 and 5 000 μmol / L groups were (14.43 ± 3.44), (22.50 ± 1.50), (37.96 ± 2.23), (59.37 ± 5.34), (93.14 ± 1.23) and (100.00 ± 1.63)%], which was significantly different from that of the control group [(7.84 ± 0.61)%] (P <0.05). In addition, the integrity and ultrastructure of V79 cells biofilm also changed. Conclusion ACN can induce V79 cell biofilm injury by altering mitochondrial membrane potential, membrane permeability and ultrastructure.
