目的:对比观察栀子、黄芩药材及栀子苷、黄芩苷对小鼠脑缺血的保护作用,探讨栀子、黄芩与其标准品及配伍防治脑缺血损伤的配伍关系。方法:采用高效液相色谱法对栀子中栀子苷和黄芩中黄芩苷进行含量测定,双侧颈总动脉结扎法复制小鼠脑缺血模型,灌胃给予栀子苷、黄芩苷及其配伍,同等含量的栀子、黄芩及其配伍药材,称量动物脑干重、湿重计算脑含水量,HE染色观察病理形态的变化。结果:每克栀子中含栀子苷为35.75毫克,每克黄芩中含黄芩苷为248毫克;含相同栀子苷的栀子药材、栀子苷(15mg/kg)和黄芩苷(15mg/kg)可以降低脑含水量,脑缺血的病理形态有改善作用。结论:栀子和黄芩的含量符合国家药典标准;栀子苷和黄芩苷对小鼠脑缺血损伤具有保护作用。
Objective: To compare the protective effect of Gardenia jasminoides, Radix Scutellariae and Geniposide and baicalin on cerebral ischemia in mice, and to explore the compatibility between Gardenia jasminoides and Radix Scutellariae and their standards and compatibility in preventing cerebral ischemia injury. Methods: The contents of geniposide and baicalin in Fructus Gardeniae were determined by high performance liquid chromatography (HPLC). The model of cerebral ischemia was induced by bilateral common carotid artery ligation in rats. Intragastric administration of geniposide, baicalin and their Compatibility, the same content of gardenia, skullcap and its compatibility herbs, weighed animal brain dry weight, wet weight calculated brain water content, HE staining observed pathological changes. Results: Each gram of gardenia contained 35.75 mg of geniposide and 248 mg of baicalin per gram of baicalin, while Gardenia jasminoides (15 mg / kg) and baicalin (15 mg / kg) can reduce brain water content, the pathological changes of cerebral ischemia have an improving effect. Conclusion: The contents of Gardenia jasminoides and Scutellaria baicalensis lie in line with the national pharmacopoeia standards. Geniposide and baicalin have a protective effect on cerebral ischemia in mice.
